Oral administration, once daily.
To facilitate administration, the required daily dose should be placed in a small amount of water and/or mixed with molasses or another sweetener and stirred until dissolved. In this case, the dissolved tablets should be administered using a syringe. The entire amount should be administered immediately.
The tablets should not be crushed.
Starting dose: The starting dose is approximately 2 µg of pergolide/kg (dose range: 1.3 to 2.5 µg/kg; see table below). The maintenance dose should be titrated according to individual response, as determined by monitoring (see below), resulting in an average maintenance dose of 2 µg of pergolide/kg body weight, with a dose range of 0.6 to 10 µg of pergolide/kg body weight. It is anticipated that lifelong treatment will be necessary for this disease.
Most horses respond to therapy and stabilize at an average dose of 2 µg of pergolide/kg body weight. Clinical improvement is expected with pergolide administration within 6 to 12 weeks. Horses may respond clinically to lower or variable doses; therefore, titration to the minimum effective dose for each animal is recommended based on the response to therapy, whether efficacy is observed or signs of intolerance appear. Some horses may require doses up to 10 µg of pergolide/kg body weight per day. In these rare situations, appropriate additional monitoring is recommended.
After the initial diagnosis, repeat endocrine tests to perform titration and monitor treatment at 4- to 6-week intervals until clinical signs stabilize or improve and/or diagnostic tests are performed.
